Preparation Alkaline Phosphatase Stabilizer

This guide describes the preparation of Alkaline Phosphatase Stabilizer at a defined molarity for laboratory use.

Alkaline Phosphatase Stabilizer is used to maintain enzyme activity during storage and handling.

Alkaline Phosphatase Stabilizer – Composition

NameFormulaConcentrationCAS
Tris-Cl (pH 9.5)C₄H₁₁NO₃100 mM77-86-1
Magnesium chlorideMgCl₂5 mM7791-18-6
Sodium chlorideNaCl100 mM7647-14-5
Tween-20C₅₈H₁₁₄O₂₆0.1% (v/v)9005-64-5

Sterilization & Storage

  • Sterilization: 0.22 μm filtration or autoclave.
  • Storage: Room temperature or 4 °C.

Tips

  • Tween-20 concentration range is 0.1–1% (v/v). Higher concentrations help reduce precipitate formation.
  • If prepared using sterile stock solutions and water, no further sterilization is required.
  • When preparing directly from anhydrous magnesium chloride, be aware of the exothermic reaction; add in small portions to partial solvent volume.
  • Using sterile stock solutions and ultrapure water eliminates the need for additional sterilization.
