Preparation Co-IP Lysis Buffer
This guide describes the preparation of Co-IP Lysis Buffer at a defined molarity for laboratory use.
Co‑IP Lysis Buffer is used for immunoprecipitation experiments to gently lyse cells while preserving protein–protein interactions.
Co‑IP Lysis Buffer – Composition
| Name | Formula | Concentration | CAS |
|---|---|---|---|
| Tris‑Cl (pH 7.4) | C₄H₁₁NO₃ | 25 mM | 77‑86‑1 |
| Sodium chloride | NaCl | 150 mM | 7647‑14‑5 |
| EDTA | C₁₀H₁₆N₂O₈ | 1 mM | 60‑00‑4 |
| NP‑40 | (C₂H₄O)nC₁₄H₂₂O | 1% (v/v) | 26027‑38‑3 |
| Glycerin | C₃H₈O₃ | 5% (v/v) | 56‑81‑5 |
Sterilization & Storage
- Sterilization: Usually not required; filtering (0.22 μm) is optional if removing particulates or microbes is desired.
- Storage: 4 °C (refrigerated; avoid long‑term storage at room temperature).
Tips
- Don’t mix concentrated stock solutions together that can cause the precipitation of salts; add buffer components to sufficient water first and then mix.
