Function of Solution III
Solution III neutralizes the alkaline lysis mixture from Solution II, precipitating chromosomal DNA, proteins, and cell debris while keeping plasmid DNA in solution.
Preparation Tips, Sterilization, and Storage
- By adding 3 M potassium acetate and 2 M acetic acid, the total acetate concentration of the solution is 5 M. Many references simply describe this as 5 M acetate, which can be misleading.
- Sterilization: autoclave.
- Storage: 4°C.
