Preparation Saline-sodium citrate (SSC) Buffer

This guide describes the preparation of Saline-sodium citrate (SSC) Buffer at a defined molarity for laboratory use.

SSC buffer is commonly used in nucleic acid hybridization washes.

Saline-sodium citrate (SSC) – Composition

NameFormulaConcentrationCAS
Sodium chlorideNaCl0.15 M7647-14-5
Sodium citrateC₆H₅O₇Na₃0.015 M68-04-2

Sterilization & Storage

  • Sterilization: Autoclave
  • Storage: Room temperature

Tips

  • 2X SSC buffer can be used for high-salt membrane washing to remove partially non-specific probe binding.
  • 0.5X SSC buffer can be used for low-salt membrane washing to increase stringency, thereby enhancing the repulsive forces between RNA and DNA strands.
